چکیده
Ca2+-free calmodulin was obtained by exhaustively dialyzing 2 mg protein (c = 4 mg/mL) against 1000 volumes of 2 mM EGTA, 50 mM Tris HCl and 150 mM NaCl (pH = 7.4) at 4 °C in a 0.5 mL Pierce Slide-A-LyzerTM Dialysis Cassette (3.5 K MWCO) for 2 days. As the carboxylic groups of EGTA exhibit strong IR bands in the region 1570-1630 cm-1 that overlap with the amide I' band of proteins (1600-1700 cm-1), EGTA was removed by subsequent dialysis against 1000 volumes MilliQ water at 4 °C for 2 days. In both cases, the dialysis buffer and water, respectively, were exchanged every 9-12 h. Calmodulin concentrationwas determined by measuring UV/Vis absorption at 276 nm with 276 = 3006 M-1 cm-1. The appropriate amount of the protein was ε aliquoted and lyophilized overnight in order to remove H2O, which is necessary for FTIR spectroscopy. All measurements were performed in D2O buffer containing 50 mM Tris HCl, 150 mM NaCl and 10 mM CaCl2 (pD 7.4), except for the apo-calmodulin experiments, in which CaCl2 was lacking in buffer. The S-tertbutylthio (StBu) protection group at the N-terminal cysteine of the synthetic peptide was removed by reduction with tris(2carboxyethyl)phosphine (TCEP) before all experiments.

متن کاملHidden connections: Network effects on editorial decisions in four computer science journals
This paper aims to examine the influence of authors’ reputation on editorial bias in scholarly journals. By looking at eight years of editorial decisions in four computer science journals, including 7179 observations on 2913 submissions, we reconstructed author/refereesubmission networks. For each submission, we looked at reviewer scores and estimated the reputation of submission authors by mea...
متن کاملThe Journal of Applied Oral Science will soon use a new online submission system
The Scientific Electronic Library (SciELO), with the support of the São Paulo Research Foundation (FAPESP), has given the opportunity for journals indexed in this database to use a more powerful online, electronic submission system. Therefore, soon the Journal of Applied Oral Science will use ScholarOne. ScholarOne has been providing scholarly publishers, societies, and associations with online...
متن کاملPREPARATION OF PAPERS FOR PAC’99{Accel Conf}
The proceedings will be produced in both electronic and paper versions{How much longer do we say paper?}. The use of electronic publishing imposes certain requirements on authors. APAC, EPAC and PAC {all accelerator conferences?} have adopted the same standards for electronic publication and have created mirrored Web sites [1], [2] for the publication of their proceedings. This document describ...
متن کاملStreamlined manuscript submission guidelines: Beyond overdue
With “high impact” medical journals having acceptance rates ranging between 5 and 11% we are all very familiar with emails similar to the one above. Such responses do not generally reflect the quality of the authors' manuscript, but have more to do with a journals focus, load and target audience. Considering that there are currently 5153 medical journals indexed in the National Library of Medic...
